It's easy to get up to speed with Posterino. If you not already did, you should watch the Getting Started screen-cast to get an idea of the basic workflow.
A general workflow looks like this:

Choose a template:

You can change the frame, filter or various other settings of each individual image by using the Inspector:

toolbar button and choose a way to export your document:Here you can choose to render your Posterino document, e.g. into an image file you can send to a print shop.
This is just a general guideline, you don't need to follow this workflow step by step. Just choose what works for you.
Generally one can say that:
Blueprint view = control the overall look of your document
Inspector = fine-tuning individual images or aspects of your document
